Chemical Fixation Analysis

These movies were taken to estimate the 'fixation times' (times for flagellar beat cessation) under a variety of common fixation conditions. In all cases fixative was added via a home-made fast fluid-exchange system. The presence of a colourant in the fixative allows a good estimate of the time at which the fixative first comes into contact with the cells. The extent of fixation can then be assessed using fast-capture videomicroscopy.
